We previously found that Nedd4-2 ubiquitinates the glutamate receptor subunit 1 (GluA1) subunit of the α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptor, which leads to reduction of neuronal excitability and seizures in mice.
We report that compared to their non-epileptic littermates, in the stargazer somatosensory cortex, there was a significant reduction in expression of AMPARs containing GluA1, 3 and 4 subunits prior to seizure onset, whereas reduction in expression of GluA2-AMPARs appears to be a post-seizure event.
We propose that insertion of GluA1 subunits at the glutamatergic synapses causes potentiation of α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic receptor (AMPAR)-mediated neurotransmission, which helps to spread and sustain seizures.

Like the rat, hypoxic seizures in P9 C57BL/6N wild-type mice resulted in transient increases in GluR1 S831 and GluR1 S845 phosphorylation in cortex and were associated with enhanced seizure susceptibility to later-life kainic-acid-induced seizures.
