The expression of TLR2, TLR4, MyD88, TRAF-6, NFκB1 and IL-8 mRNA was increased while SIGIRR, IRAK-M, A-20 and TOLLIP mRNA were decreased in fetal vs. mature human enterocytes and further altered in NEC enterocytes.

When AjTollip was co-transfected with AjMyD88 into HEK293 cells, the luciferase activities of NF-κB were significantly decreased compared with that of AjMyD88 single-transfection groups in natural state or under the stimulation of LPS and poly I:C. These results collectively suggested that AjTollip functions as a negative regulator of MyD88-dependent TLR signaling and plays an important role in fish defense against viral and bacterial infections.
