As a consequence, we found that JAK2 inhibitor AG490 inhibited cell growth and invasion, and induced cell apoptosis and cycle arrest in GBC-SD and SGC-996 cells.
Human GBC tissue and adjacent normal gallbladder tissue sections were surgically removed and miR-146b-5p expression and the development and pathological characteristics of GBC were investigated. miR-146b-5p expression was reduced in GBC tissue compared with that in adjacent tissue, and a significant correlation was observed between miR-146b-5p expression levels and carcinoma size and development. miR-146b-5p overexpression in the SGC-996 GBC cell line inhibited cell growth through enhanced apoptosis and G1 phase arrest.
In addition, using siRNA targeting TNF-α, the vector, pGPU-GFP-siTNF-α, was constructed and then transfected into the SGC-996 cells (gallbladder cancer cell line) which express high levels of endogenous TNF-α.
Furthermore, treatment of GBC lines with HDGF-targeting siRNA oligonucleotides (HDGF-siRNA) significantly reduced the proliferation of GBC-SD and SGC-996 cell lines and diminished both anchorage-independent growth on soft agar and cell migration.
