As compared to their levels in normal cells and tissues, miR-27a expression was found to be upregulated in OA cartilage and IL-1β-treated articular chondrocytes.
Primary cultured chondrocytes were challenged by IL-1β and TNF-α to simulate OA conditions. qRT-PCR was performed to detect the miR-27, lncRNA-CIR, MMP13 mRNA expression levels.
This study investigated the effects of a cyclic HP (1-5 MPa), in both normal and OA human chondrocytes, on the expression of <i>miR-27a/b</i>, <i>miR-140</i>, <i>miR-146a/b</i>, and <i>miR-365</i>, and of their target genes (<i>MMP-13</i>, <i>ADAMTS-5</i>, <i>IGFBP-5</i>, and <i>HDAC-4</i>).
There was a significant reduction (77%, p < 0.01) in miR-140 expression in OA compared to the normal chondrocytes, whereas miR-27a expression was only slightly decreased (23%).
