Viruses (previously designated group D) that contain two mutations in the activating transcription factor/cAMP response element (ATF/CRE) in the LMP-1 promoter, which are known to decrease promoter activity greatly, were significantly less frequent in Hodgkin's disease than in both infectious mononucleosis (p = 0.0081) and asymptomatic EBV carriers (p = 0.0084).

We analysed the 30-bp deletion and the single-base mutations of the LMP-1 gene in 13 spontaneously established lymphoblastoid cell lines (LCLs) from peripheral blood mononuclear cells of three healthy children, four patients with EBV-unrelated acute febrile illnesses, three patients with infectious mononucleosis (IM), and three patients with chronic active EBV infection (CEBV), and six frozen samples from four patients with CEBV and two patients with EBV-associated hemophagocytic syndrome (EBV-AHS).

Recently, focused deletions near the 3' end of the LMP-1 gene (del-LMP-1, amino acids 346-355), in a region functionally related to the half-life to the LMP-1 protein, have been reported frequently in human immunodeficiency virus (HIV)-associated HD (100%) and EBV+ Malaysian and Danish peripheral T-cell lymphomas (100%, 61% respectively), but less frequently in cases of HD not associated with HIV (28%, 33%) and infectious mononucleosis (33%).
