To study potential associations between single nucleotide polymorphisms (SNPs) in CD14, Toll-like receptor 2 (TLR2), and TLR4 genes, and atopy and new-onset asthma in young farmers.
Twenty-four single nucleotide polymorphisms (SNPs) in CD14, Toll-like receptor 4 (TLR4), TLR2, MD2 and MyD88 were genotyped in 408 agricultural workers with spirometry and questionnaire data on asthma symptoms available.
Upregulation of TLR2 and TLR4 on macrophages with increased sensitivity to hyaluronan fragments suggests a novel proinflammatory mechanism by which persistence of hyaluronan fragments could contribute to chronic inflammation and airway remodeling in asthma.
We observed that children who were heterozygous or homozygous for both the CD14/-159T and TLR2/-16934A alleles had a 4-fold lower risk for asthma than children who were carriers of the T allele of CD14 but non-carriers of the A allele of TLR2, and an almost 3-fold lower risk for asthma when compared to all other groups.
Wherease some previous studies indicated TLRs and their polymorphisms might have some role in asthma incidence and features, our data demonstrated that TLR2Arg753Gln and TLR4 Asp299Gly gene variants were not risk factors for asthma or its features in Iranian patients.
With respect to the TLR2rs4696480 polymorphism, significant association was detected between asthma susceptibility and TLR2rs4696480 polymorphism under dominant model (OR = 2.455, 95% CI = 1.235-4.88, P = .01) and codominant 3 model (OR = 2.776, 95% CI = 1.199-6.427, = 0.017).
